Homozygotes are viable and fertile with no gross physical or behavioral abnormalities. Northern blot show absence of a full length transcript in homozygous spleen and thymus. Homozygous mice exhibit prolonged bleeding time, defective platelet aggregation, and are protected from arachidonic acid induced-shock. These mice may be useful in studies of vascular biology, including hemostasis and thrombosis, as well as human TXAS-deficiency.
A targeting vector was designed to replace the distal portion exon 9 of the endogenous gene with a phosphoglycerate kinase-hypoxanthine phosphoribosyltransferase (PGK-HPRT) cassette (transcribed in the reverse orientation). This construct was electroporated into 129P2/OlaHsd-derived E14TG2a (HPRT-) embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts. Chimeric mice were bred with C57BL/6J for 5 generations, then intercrossed to homozygosity prior to arrival at The Jackson Laboratory.

Strain of Origin | 129P2/OlaHsd |
Chromosome | 6 |
Molecular Note | Sequence encoding amino acids 234-333 and the first 233 bp of intron 9 were replaced with an HPRT cassette. Southern blot and PCR using tail DNA confirmed recombination. Northern blot and RT-PCR showed lack of wild-type transcript in spleen and thymus tissues of mutants. |

When maintaining a live colony, these mice are bred as homozygotes.

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
