Description

Prdx6 knock-out mice exhibit low survival rates, severe tissue damage, and high protein oxidation levels. They may be useful to study signal transduction processes and clinically important diseases such as atherosclerosis, diabetes, and neurodegenerative diseases.

Development

A targeting vector containing the neomycin resistance and lacZ genes was used to disrupt exons 1 and 2. The construct was electroporated into R1-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts. The resulting chimeric animals were crossed once to C57BL/6J mice and maintained by sibling mating before being backcrossed for an additional three generations using a marker-assisted backcross strategy.
