These mice possess loxP sites flanking exon 2 of the neuropilin 1 (Nrp1) gene and have applications in studies related to cardiovascular and neural system development.

These mice possess loxP sites on either side of exon 2 of the targeted gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When used in conjunction with a Cre recombinase-expressing strain, this strain is useful in generating tissue-specific mutants of the floxed allele.
For example, when bred to B6.Cg-Tg(Tek-cre)1Ywa/J (Stock No. 008863) endothelial cell specific Cre expression results in mice exhibiting abnormal cardiac development and perinatal lethality.
A FRT site flanked targeting vector containing neomycin resistance and thymidine kinase genes was utilized in the construction of this mutant. loxP sites were placed flanking exon 2 and the neomycin resistance/ thymidine kinase genes. This construct was electroporated into 129.1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. The resulting chimeric mice were crossed to C57BL/6 mice and then subsequently crossed to an FLP recombinase expressing strain (B6;SJL-Tg(ACTFLPe)9205Dym/J Stock No. 003800) to remove the selection vector. Mice carrying the floxed allele were backcrossed to C57BL/6 for 10 generations (see SNP note below).
A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. While the 27 markers throughout the genome suggested a C57BL/6 genetic background, 2 of 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6J ; C57BL/6N genetic background.
