Detailed description

These NOD.IgIns-125L mice express a transgene containing the variable region of the heavy chain B-cell receptor mAB125 that binds insulin when co-expressed with a transgene encoding the light chain with the VJ of mAB125.

Development

Thomas and Hulbert received from the American Type Culture Collection (ATCC) the BALB/c IgG1 mouse anti-human insulin monoclonal antibody, mAb125 that was the source of the immunoglobulin heavy chain VDJ sequence in this transgene. This VDJ sequence was used in place of the endogenous variable sequence downstream of the Ighm sequence in the construct previously used by Goodenow et al., to generate anti-hen egg lysozyme transgenes. This new construct encodes an IgM^a constant region with the variable specificity from the mAb125 which, when co-expressed with a transgene encoding the light chain with the VJ of mAB125 (stock# 5018), binds insulin. This construct was microinjected into (B6 x D2)F1 oocytes. The transgenic founders were backcrossed to C57BL/6 more than 10 times before being backcrossed and maintained on NOD/Tac for more that 20 generations. In 2004, N25 mice were backcrossed to NOD/LtJ at The Jackson Laboratory, and are maintained hemizygote x wild type.
