Detailed description

These NOD.IgIns-125H mice express a transgene containing a light chain that binds most mammalian insulin when co-expressed with a transgene encoding the heavy chain with the VDJ of mAB125.

Development

Thomas and Hulbert received from the American Type Culture Collection (ATCC) the BALB/c IgG1 mouse anti-human insulin monoclonal antibody, mAb125 that was the source of the immunoglobulin kappa chain VJ sequence in this transgene. This VJ sequence was used in place of the endogenous variable sequence downstream of the Igkc sequence in the construct previously used by Goodenow et al., to generate anti-hen egg lysozyme transgenes. This new construct encodes a light chain that, when co-expressed with a transgene encoding the heavy chain with the VDJ of mAB125, binds most mammalian insulins. This construct was microinjected into (B6 x D2)F1 oocytes. The transgenic founders were backcrossed to C57BL/6 more than 10 times before being backcrossed and maintained on NOD/Tac for 25 generations. In 2004, N25 mice were backcrossed to NOD/LtJ at The Jackson Laboratory, and are maintained hemizygote x wild type.
