These floxed mutant mice possess loxP sites flanking exon 2 of the Ctnna1 gene. This strain may be useful for generating conditional mutations in applications where cre recombinase is expressed in cells or tissue of interest.
Elaine Fuchs, The Rockefeller University
These mice possess loxP sites on either side of exon 2 of the targeted gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities.
When bred to a strain expressing Cre recombinase in the female germline (see Stock No. 003551 for example), this mutant mouse strain may be useful in studies of mammary epithelium.
When bred to a strain expressing Cre recombinase in the mammary gland (see Stock No. 003552 for example), this mutant mouse strain may be useful in studies of mammary epithelium.
When bred to a strain expressing Cre recombinase in the nervous system (see Stock No. 003771 for example), this mutant mouse strain may be useful in studies of the cerebral cortex and the hedgehog signalling pathway.
When bred to a strain expressing tamoxifen-inducible Cre recombinase in juvenile and adult cardiac myocytes (see Stock No. 005657 for example), this mutant mouse strain may be useful in studies of dilated cardiomyopathy.
A loxP site flanked targeting vector containing a neomycin resistance gene was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 2 of the targeted gene, and another loxP site was inserted upstream of exon 2. This construct was electroporated into 129/Sv derived GS1 embryonic stem (ES) cells which were transiently transfected with a vector containing the Cre-recombinase gene under the control of the cytomegalovirus promoter to remove the selection cassette. Correctly targeted ES cells were injected into C57BL/6J blastocysts.
|Allele Name||targeted mutation 1, Elaine Fuchs|
|Allele Type||Targeted (Conditional ready (e.g. floxed), No functional change)|
|Allele Synonym(s)||alpha-cat fl; floxed alpha-E-catenin|
|Gene Symbol and Name||Ctnna1, catenin (cadherin associated protein), alpha 1|
|Strain of Origin||129X1/SvJ|
|Molecular Note||LoxP sites were inserted by homologous recombination that flanked exon 2. An adjacent floxed neomycin selection cassette was removed by Cre-mediated recombination in ES cells prior to the production of chimeric mice.|
When maintaining a live colony, homozygous mice may be bred together.
