Detailed description

Mice homozygous for the Il12rb2^tm1Jm targeted mutation are viable and fertile. Con A-activated splenocytes from homozygous mice bind IL-12 with both high and low affinity, but fail to produce IFN-gamma or proliferate in response to IL-12 stimulation; IL-12-mediated signal transduction is defective as measured by phosphorylation of STAT4. Splenocytes fail to show induced NK lytic activity when incubated with IL-12 and are deficient in IFN-gamma secretion when stimulated with either Con A or anti-CD3 mAb in vitro. Homozygous mice are deficient in vivo in their ability to produce IFN-gamma following endotoxin administration and to generate a type 1 cytokine response.

In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. This is the case for the strain above. It should be noted that the phenotype could vary from that originally described.
