Detailed description

This strain was made using gene targeting in embryonic stem cells to modify the Il4 locus by knocking-in a transmembrane domain. Expression of Il4 in a membrane-bound form provides a novel method for the identification and characterization of Il4-producing cells.

Molecular note: A modified version of exon 4 of the gene was inserted between the last codon and the translational stop site of the gene. This modified version of exon 4 contained a transmembrane anchor. In addition, a neomycin resistance cassette was inserted 150 nucleotides downstream of the polyadenylation signal of the gene. These modifications allowed production of mice that express a membrane-bound form of the protein.
General note: This mutation resulted in mice that express Il4 in a membrane-bound form for use in identifying and characterizing Il4-producing cells.
