Detailed description

Mice homozygous for the Col3a1^tm1Jae targeted mutation exhibit a shorter life span with approximately 10% of homozygous mutant mice surviving to adulthood. The major cause of death of is rupture of the major blood vessels.

Development

The first two exons of the Col3a1 were replaced by a vector containing a neo resistance gene. The construct was electroporated into 129S4/SvJae-derived J1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to C57BL/6. These mice were backcrossed to BALB/c for at least 5 generations.
