Mice homozygous for Icam1tm1Jcgr lack surface Intercellular adhesion molecule 1 (ICAM-1) expression with elevated numbers of circulating neutrophils and lymphocytes, as well as diminished allogeneic T cell responses and delayed type hypersensitivity. The phenotype is similar to C57BL/6J-Icam1tm1Bay (Stock No. 002127) but are reported to be less leaky.

Mice homozygous for the Icam1tm1Jcgr targeted mutation are viable and fertile. The phenotype is similar to C57BL/6J-Icam1tm1Bay (Stock No. 002127) but are reported to be less leaky. Characteristics include longer lifespan and nephritis with no apparent lung involvement.
In 2018, single nucleotide polymorphism (SNP) panel analysis suggested Stock No. 002867 has the Cdh23 functional allele (Cdh23+) normally found for 129 mice rather than the age related hearing loss mutant allele (Cdh23ahl) normally present for C57BL/6 mice. Additionally, if this colony is bred to C57BL/6J inbred mice after 2018, it may then be segregating for either Cdh23ahl or Cdh23+. See strain Development section if any additional details are known.
This strain was generated by insertion of a vector containing the neo resistance gene into exon 4 of the Icam1 gene on chromosome 9. The 129-derived J1 ES cell line was used.
In 2018, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the living colony (generations N10 and N11) at The Jackson Laboratory Repository. This revealed 46/48 markers to be C57BL/6J allele-type (including all 5 markers that determine C57BL/6 substrains). All mice tested had two markers homozygous for 129 allele-type - one on chromosome 4 [~20 Mbp] and one on chromosome 10 [~60 Mbp]. Because the chromosome 10 SNP marker lies within the Cdh23 locus, these data suggest Stock No. 002867 has the Cdh23 functional allele (Cdh23+) normally found for 129 mice rather than the age related hearing loss mutant allele (Cdh23ahl) normally present for C57BL/6 mice.
Additional note: If this colony is bred to C57BL/6J inbred mice after 2018, it may then be segregating for either Cdh23ahl or Cdh23+.
|Allele Name||targeted mutation 1, Jose Carlos Gutierrez Ramos|
|Allele Type||Targeted (Null/Knockout)|
|Allele Synonym(s)||ICAMtm1-jcgr; ICAM-1-; ICAM-1-deficient|
|Gene Symbol and Name||Icam1, intercellular adhesion molecule 1|
|Strain of Origin||129S4/SvJae|
|Molecular Note||A neomycin resistance cassette was inserted into exon 4 of the gene. While King, et. al. (J:25530) reported the potential for the production of functional proteins arising from alternative transcripts, corresponding signals were undetected by immunohistochemical analysis of homozygous mutant mice.|
