Description

In this strain, XX mice that carry the Tg(Sry)2Ei transgene are sex-reversed males.

Detailed description

This is one of three transgenic strains carrying the Tg(Sry) transgene (the Y-linked Sry gene derived from a 129 inbred strain) on a C57BL/6 background with an AKR Y chromosome. All XX mice are sex-reversed males. (See also Stock No. 002063) and Stock No. 002837).

Development

The 129-derived 14.6-kb of genomic sequence encoding Sry, described by Gubbay et al. (Nature, 1990. v346:245), was injected into the male pronuclei of fertilized eggs obtained from mating C57BL/6JEi females with C57BL/6JEi-Chr Y^AKR/J males. Three XY transgenic male founders were obtained and each generated a line that was maintained by backcrossing transgenic males, obligatorily carrying the consomic Chr Y^AKR/J, to C57BL/6JEi females (Eicher et al., 2001). All three strains were independently cryopreserved. The cryo-recovered males have the Chr Y^AKR/J and those that additionally carry Tg(Sry)2Ei should be used as the breeders to maintain the line. The cryo-recovered females will be either lack Tg(Sry)2Ei and be wild-type or carry Tg(Sry)2Ei and be sex reversed sterile phenotypic males. These embryos were generated from carrier males bred to C57BL/6J females, so are no longer pure C57BL/6JEi in the autosomes, but rather an F1 hybrid mix of C57BL/6JEi and C57BL/6J.
