Mice homozygous for the Cd8a^tm1Mak targeted mutation are deficient in functional cytotoxic T-cells; however, helper T-cell development and function is comparable to normal.

Development

A targeting vector containing neomycin resistance gene was used to disrupt exon 1. The construct was electroporated into 129S2/SvPas derived D3 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts.

The resulting chimeric animals were crossed to C57BL/6 mice. Heterozygotes were intercrossed to generate homozygotes. The mice were then backcrossed to C57BL/6 for 13 generations before arriving at The Jackson Laboratory.
