Macrophages from mice homozygous for the Ifngtm1Ts knock-out allele show impaired production of antimicrobial products and reduced expression of MHC II antigens. There is also uncontrollable proliferation of splenocytes from homozygotes in response to mitogens and alloantigens and a reduced resting natural killer cell activity.

Mice homozygous for the Ifngtm1Ts targeted mutation are viable and fertile. Homozygotes appear normal in a "clean" environment but display reduced macrophage function in response to pathogens. Specifically, macrophages show impaired production of antimicrobial products and reduced expression of MHC II antigens. There is also uncontrollable proliferation of splenocytes from homozygotes in response to mitogens and alloantigens and a reduced resting natural killer cell activity.
A targeting vector containing a neo cassette was used to disrupt exon 2. The construct was electroporated into 129S7/SvEvBrd-Hprt1+ derived AB1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting mice were backcrossed onto the C57BL/6 background for 8 generations by the donating investigator before arriving at The Jackson Laboratory.
