Description

Mice homozygous for the Ifng^tm1Ts targeted mutation appear normal in a "clean" environment but display reduced macrophage function in response to pathogens. Specifically, macrophages show impaired production of antimicrobial products and reduced expression of MHC II antigens. There is also uncontrollable proliferation of splenocytes from homozygotes in response to mitogens and alloantigens and a reduced resting natural killer cell activity.

Detailed description

Mice homozygous for the Ifng^tm1Ts targeted mutation are viable and fertile. Homozygotes appear normal in a "clean" environment but display reduced macrophage function in response to pathogens. Specifically, macrophages show impaired production of antimicrobial products and reduced expression of MHC II antigens. There is also uncontrollable proliferation of splenocytes from homozygotes in response to mitogens and alloantigens and a reduced resting natural killer cell activity.

Development

A 2-kb neomycin-resistance gene (neo) was inserted into exon 2, which introduced a termination codon after the first 30 amino acids of the mature IFN-y protein. The targeting vector was transfected into AB-1 embryonic stem cells. Injection of the selected 48D clone with a disrupted allele into C57BL/6J blastocysts generated chimeric mice and heterozygous offspring were intercrossed to produce homozygotes for the targeted mutation of the IFN-y gene. The strain was backcrossed onto a BALB/c background for 6 generations by the donating investigator before the strain was imported to IMR.
