Description

These C3H.SW mice carry a Y Chromosome from the SWR/J strain with males developing extreme hyperinsulinemia and extreme islet hyperplasia.

Detailed description

C3.SW-H2^b/SnJ (Stock number 000438) males exhibit maturity onset impairment of glucose tolerance and hyperinsulinemia. In contrast to Stock number 002110, this strain carries the C3.SW-H2^b/SnJ Y chromosome. Males develop extreme hyperinsulinemia, transiently elevated plasma glucose and extreme islet hyperplasia. This strain and Stock number 002110 demonstrate that the Y chromosome modulates the level of hyperinsulinemic and glycemic stress exerted upon pancreatic beta cells.

Development

Congenic C3HeB/FeChp males carrying the obesity gene, Lepr^db (chromosome 4) were backcrossed to C3.SW-H2^b/SnJ females to produce C3.SW-H2^b/Lt mice heterozygous for Lepr^db. F1 mice were intercrossed, to trap H2^b homozygotes in the F2 generation. The C3.SW-H2^b/SnJ Y chromosome was introduced by reciprocal crosses in the first backcross. At the second backcross Lepr^dbwas eliminated. The colony has been backcrossed eight times to C3.SW-H2^b/SnJ then intercrossed for 21 generations. The frozen stock is N9.
