This is one of three transgenic strains carrying the Tg(Sry) transgene (the Y-linked Sry gene derived from a 129 inbred strain) on a C57BL/6 background with an AKR Y chromosome. All XX mice are sex-reversed males. (See also Stock No. 002708) and Stock No. 002837).
XYAKR/J mice heterozygous for TOrl on a C57BL/6J background develop as sex reversed females, but in the presence of the Tg(Sry)1Ei transgene these XY mice develop as normal males with testes.
The 129-derived 14.6-kb of genomic sequence encoding Sry, described by Gubbay et al. (Nature, 1990. v346:245), was injected into the male pronuclei of fertilized eggs obtained from mating C57BL/6JEi females with C57BL/6JEi-Chr YAKR/J males. Three XY transgenic male founders were obtained and each generated a line that was maintained by backcrossing transgenic males, obligatorily carrying the consomic Chr YAKR/J, to C57BL/6JEi females (Eicher et al. 2001). All three strains were independently cryopreserved. The cryo-recovered males have the Chr YAKR/J and those that additionally carry Tg(Sry)1Ei should be used as the breeders to maintain the line. The cryo-recovered mice with two X Chromsosomes will be either lack Tg(Sry)1Ei and be wild-type or carry Tg(Sry)1Ei and be sex reversed sterile phenotypic males.

General Note | Three transgenic lines (referred to as TgN(Sry-129)1Ei, TgN(Sry-129)2Ei, and TgN(Sry-129)3Ei were generated which all produced sex-reversed XX transgenic male offspring. No copy number or insertion location information was provided. |
Molecular Note | This transgene consists of an inbred 129 strain derived 14.6 Kb genomic fragment containing the entire Sry open reading frame and no other genes. |

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
