Description

These T(16;17)43H trisomic mice exhibit spatial learning deficits.

Detailed description

T43H encompasses >310 known genes triplicated within the proximal 30 megabases (Mb) of Chr17. The Ts43H mice are viable on a mixed genetic background and exhibit spatial learning deficits analogous to those observed in Ts65Dn mice with unrelated trisomy. Quantitative analysis of the brain expression of 20 genes inside the trisomic interval and 12 genes lying outside on Chr17 revealed 1.2-fold average increase of mRNA steady-state levels of triplicated genes and 0.9-fold average down-regulation of genes beyond the border of trisomy.

Development

This stock was previously maintained homozygous for the reciprocal translocation T(16;17)43H with one copy of brachyury (T) in coupling on one Chr 17. Brachyury is homozygous lethal but provides a dominant marker of the presence of the translocation-bearing chromosome, evident by a short tail. Thus, breeding together short tailed T(16;17)43H +/T(16;17)43H T translocation homozygotes produces more short tailed T(16;17)43H +/T(16;17)43H T translocation homozygotes and normal tail T(16;17)43H +/T(16;17)43H + translocation homozygotes. This cryopreserved bankstock of embryos was generated by breeding C57BL/6JEi females with T(16;17)43H +/T(16;17)43H T males so the cryorecovered pups are expected to all be heterozygous for T(16;17)43H and some will be additionally heterozygous for brachyury. No molecular genotyping assay is available for this translocation. Researchers will be provided the cryorecovered mice and are encouraged to karyotype the mice received. Molecular note: This chromosome is a result of a reciprocal translocation with one breakpoint located within the centromeric heterochromatin of Chr16, and the other located in the B3 band of Chr17. In conjunction with a normal Chr16 and two normal Chr17s, the presence of this mutation results in segmental trisomy of the proximal 30 Mb of Chr17, which includes over 310 known genes, and monosomy for the tip of centromeric heterochromatin of Chr16.
