Detailed description

These I-FAB^{-277 to +28}/hGH⁺³ mice express a transgene containing the human growth hormone sequence controlled by the rat intestinal fatty acid binding protein promoter. These mice exhibit inappropriate expression of hGH in proliferating and nonproliferating epithelial cells located in the mid and upper portions of duodenal, jejunal, ileal, and colonic crypts without affecting the "shape" of the cephalocaudal gradient of transgene expression.

Development

Nucleotides -277 to +28 of the promoter of rat intestinal fatty acid binding protein were obtained as a BstEII-EcoRI fragment and fused to nucleotide +3 of the human growth hormone sequence. This was microinjected into (C57BL/6J x C3H) F1 oocytes and the offspring were backcrossed once then intercrossed. Details of subsequent breeding prior to cryopreservation are not known. This line derived from founder #54. (Cohn et al., 1992.)
