Detailed description

These I-FAB^{-184 to +28}/hGH⁺³ mice express a transgene containing the human growth hormone sequence controlled by the rat intestinal fatty acid binding protein promoter. Expression of the transgene occurs in replicating crypt epithelial cells but is ultimately restricted to the enterocytic lineage in both small intestine and colon.

Development

Nucleotides -184 too +28 of the promoter of rat intestinal fatty acid binding protein were obtained as a BstEII-EcoRI fragment and fused to nucleotide +3 of the human growth hormone sequence. This was microinjected into (C57BL/6J x C3H) F1 oocytes and the offspring were backcrossed once then intercrossed. Details of subsequent breeding prior to cryopreservation are not known. This strain derived from founder #39 which was reported to have integrated only 1 to 2 copies of the transgene. (Cohn et al., 1992.)
