This stock carries a radiation induced lethal mutation made from mice carrying the In(1)1Rk inversion (Stock No. 000716). The lethal mutation in I1Rk2 may act at birth and possibly after implantation. It was not determined if the mutation was located within the inverted segment (Roderick, 1983).
A male homozygous for the inversion In(1)1Rk was exposed to 1000 rads from a cesium irradiator and subsequently bred to a female heterozygous for splotch and homozygous for leaden (Pax3Sp Mlphln/Pax3+ Mlphln). Splotch and leaden are within the inverted segment in In(1)1Rk and thus do not recombine when in repulsion with this inversion. The offspring that expressed the splotch mutation were thus heterozygous for splotch and leaden in repulsion with In(1)1Rk and any radiation-induced recessive lethal mutation. When sibling intercrossed, these splotch expressing repulsion heterozygotes generate embryos homozygous for splotch, which are in utero lethal, homozygous for any induced recessive mutation, and the same genotype as the parental animals, Pax3Sp Mlphln/In(1)1Rk l1Rk2. This is how the l1Rk2 mutation was induced, isolated, and maintained prior to cryopreservation. Embryos were generated for cryopreservation from Pax3Sp Mlphln/In(1)1Rk l1Rk2 males bred with either Pax3Sp Mlphln/Pax3+ Mlphln or Pax3+ Mlphln/Pax3+ Mlphln females. The cryorecovered pups that do not have the leaden dilution of the coat color but do have the spots of the splotch mutation can be expected to be carriers of the recessive lethal l1Rk2 mutation, with the exception of less than one percent; nine crossover events were observed in 1207 mice. The recovered line can be maintained by intercrossing these carriers.

Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
