Test matings failed to produce homozygotes, indicating that this is a recessive embryonic lethal mutation. No abnormal phenotype has been reported in the heterozygote.

Development

The recessive lethal Gpi1^a-m1Ehs allele was induced by triethylenemelamine treatment of a DBA/2J male, which was crossed to a C57BL/6J (Gpi1^b) female (Soares 1979). This mutation was imported from Dr. E. R. Soares into the laboratory of Dr. Eva Eicher at The Jackson Laboratory in 1979 and was maintained by breeding to C57BL/6J and DBA/2J in alternating generations. A Gpi1^a-m1Ehs/Gpi1^a male was imported and mated to a C57BL/6J female, then the Gpi1^b/Gpi1^a-m1Ehs offspring were mated to BALB/cBy (Gpi1^a) and their Gpi1^a-m1Ehs/Gpi1^a offspring were bred to C57BL/6J. Compound heterozygotes were screened by cellulose acetate gel electrophoresis to determine genotype. This system of breeding continued until 1982 when embryos were generated for cryopreservation by breeding Gpi1^a/Gpi1^c males with C57BL/6J females. The cryorecovered pups will be a mix of Gpi1^b/Gpi1^c and Gpi1^b/Gpi1^a. To continue the line, the Gpi1^b/Gpi1^c animals should be bred to BALB/cByJ (Gpi1^a) and their Gpi1^a/Gpi1^c offspring should then be bred to C57BL/6J.

Molecular note: This allele determines a faster migrating variant of the encoded enzyme and occurs in the wild and in strains BUB/Bn, CBA/J, CBA/CaJ, C3H/HeH, C3Heb/FeJ, C57BL/6J, C57BL/6J-ob, SEA/GJ and SWR/J. Sequencing analysis identified four nucleotide differences between this allele and Gpi1^a. Two differences were silent, while a third difference at nucleotide position 282 predicts an aspartate for the a allele and an asparagine for the b allele at amino acid position 94. A fourth difference in the 3' untranslated region was also noted.
