Development

The T(7;15)9H translocation was induced prior to 1964 in a (C3H/HeH x 101/H)F1 male by exposure to 600R X-rays at 6 weeks of age and again at 14 weeks of age. Twelve weeks after the last exposure the male was bred to a CBA/H female and the female offspring of this cross were bred to C57BL/H males. Subsequent breeding is not known until this translocation was imported into the laboratory of Dr. Eva Eicher at The Jackson Laboratory in the mid-1970?s. It was bred to (C57BL/6J-A^w-J/J x CBA/CaGnLeJ)F1 mice for 7 generations. The Gpi1^a allele co-segregates with the translocation and C57BL/6J mice have the Gpi1^b allele. Thus, Gpi1^a was used as a marker for the likely presence of the translocation in breeder selection. This strain was later intercrossed to homozygosity and maintained by sibling inbreeding translocation homozygotes. In 1983 homozygous males, then at generation N7F8, were bred t o C57BL/6J females to generate embryos for cryopreservation.

Molecular note: This allele determines a faster migrating variant of the encoded enzyme and occurs in the wild and in strains BUB/Bn, CBA/J, CBA/CaJ, C3H/HeH, C3Heb/FeJ, C57BL/6J, C57BL/6J-ob, SEA/GJ and SWR/J. Sequencing analysis identified four nucleotide differences between this allele and Gpi1^a. Two differences were silent, while a third difference at nucleotide position 282 predicts an aspartate for the a allele and an asparagine for the b allele at amino acid position 94. A fourth difference in the 3' untranslated region was also noted.
