Description

This C57BL/10ScSn congenic strain carries both H46^b and Selenos^b (associated with the previously identified H47 antigen), along with the pink-eyed dilution mutation, in a single congenic interval from 129/Rr.

Detailed description

The 21M congenic interval in this strain, originally named B10.129(21M) by Snell and Stevens (1961), was initially defined as containing a histocompatibility locus that caused rejection of C57BL/10ScSn derived tumor cells by the congenic. This histocompatibility locus was designated histocompatibility 4 (H4), and was found to be closely linked to the pink-eyed dilution locus (Oca2) on Chromosome 7. It was subsequently determined by Davis and Roopenian (1989) that this H4-containing interval could be separated into two separate minor histocompatibility loci, designated H46 and H47, with H46 proximal of Oca2 and H47 distal. Further, H46 was found to be recognized by CD4+ cells whereas H47 is restricted to CD8+ cells. Mendoza et al (2001) determined that the underlying molecular cause for the H47 histoincompatibility is a single base pair transversion, A to T, that results in the substitution of phenylalanine in place of isoleucine at amino acid residue 119 in the transmembrane domain of selenoprotein S, an oxidoreductase of the endoplasmic reticulum. As these discoveries were made, the strain name evolved from B10.129(21M) to B10.129-H4^b/Sn and later B10.129P2-H46^b H47^b/Sn before the molecular variation of H47^b was characterized and assigned to a gene symbol. Because the pink-eyed dilution mutation carried along in this congenic interval from 129/Rr, the homozygotes are of a pallid-type color.

Development

Strain B10.129(21M) was derived from an initial cross between C57BL/10ScSn and 129/Rr (antecedent of the current 129P3/J, Stock No. 000690) and developed into a congenic strain by a cross-intercross system resulting in the equivalent of 11 generations of backcross to C57BL/10ScSn before being maintained by sib matings. The strain was initially identified as carrying the H4^b antigenic allele, which was later shown to consist of two linked histocompatibility genes, H46 and H47. Embryos were cryopreserved in 1994.

Oca2^p is a very old mutation carried in many varieties of fancy mice (J:12958). It has been suggested that the original mutation occurred in Japanese wild mice, Mus musculus molossinus (J:19782).

The 2 nucleotide sequence variations underlying this are G73A and C74T and the core antigenic peptide from each compete similarly for binding A^b. The core antigenic peptide in H46-b is HMFVEAIPELQ.
