Detailed description

This B6/By congenic strain carries the ‘minor’ histocompatibility allele, H22^c, along with the Gpi1^a allele from BALB/cBy. The Gpi1 congenic mice are used as markers in transplant studies.

Development

Dr. Donald Bailey, then at University of California Medical Center in San Francisco, began to generate a panel of congenic lines with differential loci that determine histocompatibility by crossing BALB/cBy females with C57BL/6By males, and then backcrossing at each generation to a C57BL/6J male. At backcross generation 5 orthotopic tailskin graft tests were done to identify lines bearing BALB/cBy-derived congenic intervals with histocompatibility loci. This backcrossing, with testing, was continued until siblings at backcross generation 11 or more were intercrossed to breed the congenic interval to homozygosity and each strain was maintained by sibling inbreeding (Bailey, 1975). Dr. Bailey transferred the mice to The Jackson Laboratory when he joined the staff in 1967. The strain was cryopreserved in 1981-82.

Molecular note: This allele determines a slower migrating variant of the encoded enzyme and occurs in the wild and in strains 101/H, A/HeJ, AKR/J, BALB/cJ, C57BR/cdJ, C58/J, CE/J, DBA/2J, DE/J, DW/J, LP/J, MA/J, NZB, PL/J, P/J, RARF, RF/J, SJL/J, ST/bJ, and 129/J. Sequencing analysis identified four nucleotide differences between this allele and Gpi1^b. Two differences are silent, while a third difference at position nucleotide position 282 predicts an aspartate for the a allele and an asparagine for the b allele at amino acid position 94. A fourth difference in the 3' untranslated region was also noted.
