Slaty, a mutation at the dopachrome tautomerase locus (Dctslt), arose spontaneously in the non-inbred balanced stock of limb deformity Jackson and mahogany (Grem1ld-J +/+ Atrnmg) in 1963. The first slaty male was outcrossed to a C57BL/6J female and the stock was sibling mated. A slaty male at F12 was outcrossed to a CBA/Ca female in 1967 and the stock was sibling mated to F9. An F9 slaty male was crossed to a C57BL/6J female and the slaty mutation was continually backcrossed to C57BL/6J by the backcross-intercross system to N10 at which time sibling matings were again used. It was cryopreserved by mating heterozygous females with homozygous males and vice versa at generation N10F49.
|Gene Symbol and Name||Dct, dopachrome tautomerase|
|Strain of Origin||STOCK Grem1ld-J Atrnmg|
|Molecular Note||Sequence analysis showed this allele involves a G-to-A transition (C-to-T on forward strand) at coding nucleotide 581 which results in a arginine to glutamine missense mutation at position 194 (p.R194Q). The mutation reduces but does not eliminate enzyme activity.|
