Notes

This assay does not work well without the use of a Hotstart Taq.

Only the two primers 10987/10988 are necessary for genotyping  the loxp flanked allele in Stock#  013167.  Adding primer 10989 (mutant  primer for null strain Stock# 013168) will not confuse loxp fanked genotyping.
The genotyping protocol(s) presented here have been optimized for reagents and conditions used by The Jackson Laboratory (JAX). To genotype animals, JAX recommends researchers validate the assay independently upon receipt of animals into their facility. Reaction cycling temperature and times may require additional optimization based on the specific genotyping reagents used.

Expected Results

Mutant (loxp flanked) = 399 bp
Heterozygote = 399 bp and 297 bp
Wild type = 297 bp

JAX uses a very high speed Taq (~1000 bp/sec), use cycling times recommended for your reagents.
