Protocol 28936: Separated PCR Assay - Scn1a<tm1.#Dsf>
Version 1.2

Notes

The genotyping protocol(s) presented here have been optimized for reagents and conditions used by The Jackson Laboratory (JAX). To genotype animals, JAX recommends researchers validate the assay independently upon receipt of animals into their facility. Reaction cycling temperature and times may require additional optimization based on the specific genotyping reagents used.

Expected Results

Homozygote Post-Cre= 198
Homozygote Pre-cre = 198 and ~410
Heterozygote Post-Cre = 164 bp and 198 bp
Heterozygote Pre-Cre = 164 bp and 198 bp and ~410 bp
Wild type = 164 bp

This assay can be used from both the Scn1a<tm1.1Dsf> (Pre-cre) and Scn1a<tm1.2> (Post-cre)

410 bp band is showing removal of Neo after FLP mediated recombination.

JAX uses a very high speed Taq (~1000 bp/sec), use cycling times recommended for your reagents.
JAX uses a 'touchdown' cycling protocol and therefore has not calculated the optimal annealing temperature for each set of primers.
