Uoxtm1Bay Allopurinol Therapy
Therapy necessary for homozygous breeding units and their wean or segregating litters from heterozygous breeder pairs.
- Obtain allopurinol from Sigma Chemicals #A8003 25 gm bottle - $60.00.
- Send a sample of each chemical lot to Diagnostic for microbe assay (routine procedure at The Jackson Laboratory)
- Measure out 150 mg allopurinol. Please following warning label on bottle: may sensitize skin; should not be inhaled or swallowed.
- Fill to 1 liter (1000ml) with water from bottles. Final concentration 0.15 mg/ml.
- Close container and shake to mix. Refill bottles for mice. To conserve useage fill bottles - 3/4 full. Water should be yellowish so bottles containing the mixture can be identified from regular water bottles. if there is a bottle run-out over the weekend, a regular bottle can be used as replacement--note this for the attending caretakers--and put back on allupurinol as soon as possible the following week.
- Store unused allopurinol solution in the refrigerator. Make up only as needed because 1) light breaks down the chemical and 2) chemical loses activity over time.
Note: There is a range of effective concentrations. Please refer to the following reference for more information.
